Despite having only a 30-minute warm-up hit together, Murray formed an instant understanding on No2 Court with Hingis, who said it was she who suggested the pairing.
'He's too nice a guy to approach so I had to take the first step,' Hingis said with a smile.
Murray won the mixed doubles with Jelena Jankovic 10 years ago but had focused on the men's doubles recently.
But he and partner Bruno Soares were knocked out on Friday night and Murray was thankful to have another reason to stay.
It is 20 years since Hingis won her first Wimbledon singles title aged 16 and she has also won three doubles titles at the All England Club, most recently in 2015 when partnering India's Leander Paes.
'I'm relieved,' said Hingis. 'I was saying to Jamie, even 20 years later I still get nervous. But I guess it's a good thing, the adrenaline's pumping. From the first to the second set it's already better. The understanding is there.'
Hingis quipped of the time Murray won the mixed: 'I've been playing for 20 years, 10 is nothing.' Murray replied: 'I've got some catching up to do.'
Murray and Hingis have been installed as No1 seeds. British duo Marcus Willis and Jay Clarke sparked jubilation on No3 Court by pulling off a late-evening victory over reigning men's doubles champions Nicolas Mahut and Pierre-Hugues Herbert. They won 3-6, 6-1, 7-6 (7-3), 5-7, 6-3.
